The material of the Speaker Surround isn't coated in any way so permanent discouloration should only occour when in contact with an aggressive agent.
In that case it should be possible to remove with a little water and dish soap. Alternatively a mild plastic cleaner should work.
Make sure to use a lint-free and damp-only cloth.

It is not a problem to use a paint brush to clean of dust off the Woofer membrane. Just be aware that in a Coax-Speaker, the tweeter sits right in the middle of the cone, instead of a dust cap. Pushing the tweeter in can, of course, damage it permanently.

Generally, you can use compressed air to clean the tweeter of dust-particles, but there is a risk of damaging the tweeter due to too high air pressure.
With these kind of manual blowers it is not possible to direct and control the force of air pressure enough to avoid pushing the tweeter dome inward.
Pressured air in a can would suit the task better, but a thin paint brush would be the safest option due to better control over force excerted.
In any case, never direct the air directly onto the tweeter dome, instead move to the sides of the dome.
